Preparation and Evaluation of Polymer Fused Metformin Hydrochloride Sustained Release Tablet

Abstract: Introduction: Diabetes Mellitus (DM) is one of the most prevalent and chronic illnesses associated with an abnormally high level glucose in body which hazardous to organs. Metformin Hydrochloride (HCl), a biguanide derivative used for Type 2 DM. It has relatively short plasma half-life its regular administration required maintain normal blood diabetic patients. Therefore, sustained-release medications are suitable approach increase patient compliance extend duration effect metformin 8-12 hr. The main goal this investigation was prepare oral tablet HCl using natural polymers as release rate-controlling agents. Materials Methods: Polymer fused hydrochloride formulated sodium alginate, pectin alone combinations at different ratios, by direct compression method. Evaluations: formulation evaluated pre-compression parameters, like drug-excipient interaction (using Fourier-transform infrared spectroscopy), drug solubility, flow properties density powder blends. compressed tablets were diameter, friability, thickness, weight variation, hardness, content uniformity vitro release. Results: study showed that alginate combination able sustain also suggested if amount polymer increased, decreased. Formulation (MA3) containing highest (250mg) gave 93.06% after 12 hr therefore chosen best formulation.
